2001 journal article

Leaky Lactococcus cultures that externalize enzymes and antigens independently of culture lysis and secretion and export pathways

APPLIED AND ENVIRONMENTAL MICROBIOLOGY, 67(1), 251–259.

By: S. Walker n & T. Klaenhammer n

MeSH headings : Antigens, Bacterial / metabolism; Bacterial Proteins / genetics; Bacterial Proteins / metabolism; Bacteriophages / enzymology; Bacteriophages / genetics; Bacteriophages / physiology; Culture Media; Genetic Engineering; Lactococcus lactis / enzymology; Lactococcus lactis / genetics; Lactococcus lactis / growth & development; Lactococcus lactis / metabolism; Lysogeny; N-Acetylmuramoyl-L-alanine Amidase; Viral Proteins / genetics; Viral Proteins / metabolism; Virus Integration; beta-Galactosidase / metabolism
TL;DR: A novel system that leaks β-galactosidase (β-gal) without a requirement for secretion or export signals was developed in lactococcus lactis by controlled expression of integrated phage holin and lysin cassettes, pointing toward the development of bacterial strains able to efficiently release relevant proteins and enzymes outside the cell in the absence of known secretion and export signals. An explosive antisense RNA strategy for inhibition of a lactococcal bacteriophage

APPLIED AND ENVIRONMENTAL MICROBIOLOGY, 66(1), 310–319.

By: S. Walker n & T. Klaenhammer n

MeSH headings : Bacteriophages / genetics; Bacteriophages / physiology; Cloning, Molecular; Escherichia coli / genetics; Gene Expression Regulation, Viral; Lactococcus / virology; Open Reading Frames / genetics; Plasmids / genetics; Polymerase Chain Reaction; Promoter Regions, Genetic; RNA, Antisense / metabolism; RNA, Messenger / metabolism; Replication Origin; Sequence Analysis, DNA; Transformation, Bacterial; Virus Replication
TL;DR: Growth curves performed in the presence of varying levels of phage φ31 showed that ori31plus anti-ORF 4H offered significant protection to Lactococcus lactis, even at multiplicities of infection of 0.01 and 0.1. (via Semantic Scholar)

A triggered-suicide system designed as a defense against bacteriophages

JOURNAL OF BACTERIOLOGY, 179(21), 6741–6748.

MeSH headings : Amino Acid Sequence; Bacteriophages / genetics; Bacteriophages / growth & development; DNA Restriction-Modification Enzymes / genetics; DNA Restriction-Modification Enzymes / metabolism; Genetic Engineering; Industrial Microbiology / methods; Lactococcus lactis / virology; Molecular Sequence Data; Site-Specific DNA-Methyltransferase (Adenine-Specific) / genetics; Site-Specific DNA-Methyltransferase (Adenine-Specific) / metabolism
TL;DR: A novel bacteriophage protection system for Lactococcus lactis based on a genetic trap, in which a strictly phage-inducible promoter isolated from the lytic phage phi31 is used to activate a bacterial suicide system after infection, was developed. (via Semantic Scholar)
