EGF receptors of hepatocytes from rats treated with phenobarbital are sensitized to down-regulation by phenobarbital in culture

TOXICOLOGY AND APPLIED PHARMACOLOGY, 165(2), 115–126.

By: . Dalton n, R. Jirtle* & S. Meyer n

author keywords: epidermal growth factor receptor; phenobarbital; CYP2B1/2; tumor promotion
MeSH headings : Animals; Aryl Hydrocarbon Hydroxylases; Barbiturates / pharmacology; Blotting, Northern; Blotting, Western; Carcinogens / pharmacology; Cells, Cultured; Cytochrome P-450 CYP2B1 / biosynthesis; Cytochrome P-450 Enzyme System / biosynthesis; Down-Regulation / drug effects; Enzyme Induction / drug effects; ErbB Receptors / drug effects; ErbB Receptors / metabolism; Female; Liver / cytology; Liver / drug effects; Liver / metabolism; Liver Neoplasms, Experimental / chemically induced; Liver Neoplasms, Experimental / pathology; Male; Phenobarbital / blood; Phenobarbital / pharmacology; Prohibitins; RNA / biosynthesis; RNA / isolation & purification; Rats; Rats, Inbred F344; Sodium-Potassium-Exchanging ATPase / metabolism; Steroid Hydroxylases / biosynthesis
TL;DR: A role for impaired EGFr function in PhB liver tumor promotion due to effects on existing EGFr protein is supported and it is suggested that EGFr down-regulation by PhB in culture is independent of CYP2B1/2 activity but shares mechanistic components involved in its transcriptional activation byPhB. Evidence for site-specific bioactivation of alachlor in the olfactory mucosa of the Long-Evans rat

TOXICOLOGICAL SCIENCES, 49(2), 202–212.

By: B. Wetmore n, A. Mitchell, S. Meyer* & M. Genter*

author keywords: mutagenicity; nasal carcinogenicity; cell proliferation; olfactory mucosa; mouse lymphoma assay; alachlor
MeSH headings : Acetamides / metabolism; Animals; Biotransformation; Blotting, Western; Body Weight / drug effects; Cell Division / drug effects; Dose-Response Relationship, Drug; Herbicides / metabolism; Lymphoma / etiology; Male; Mice; Mutagenicity Tests; Mutagens / metabolism; Mutagens / toxicity; Mutation; Olfactory Mucosa / drug effects; Olfactory Mucosa / metabolism; Olfactory Mucosa / pathology; Rats; Rats, Long-Evans; Salmonella typhimurium / genetics; Time Factors
TL;DR: Alachlor was administered i.p. to male Long-Evans rats for up to 28 days at doses that are carcinogenic in chronic studies in order to study olfactory lesion development and alterations in cell proliferation, suggesting that target tissue bioactivation of alachlor results in the formation of one or more mutagenic metabolite(s), which may be critical in alACHlor-induced nasal tumorigenesis. By: S. Meyer*, T. Kim, G. Moser, N. Monteiroriviere & R. Smart*

Contributors: S. Meyer*, T. Kim, G. Moser, N. Monteiroriviere & R. Smart*

MeSH headings : Adenine / physiology; Animals; Base Sequence; Carcinogens / toxicity; Dose-Response Relationship, Drug; Drug Synergism; Female; Genes, ras / drug effects; Genes, ras / genetics; Mice; Mice, Inbred Strains; Mirex / toxicity; Molecular Sequence Data; Mutation / drug effects; Mutation / genetics; Papilloma / chemically induced; Papilloma / genetics; Skin / drug effects; Skin Neoplasms / chemically induced; Skin Neoplasms / genetics; Tetradecanoylphorbol Acetate / toxicity; Thymidine / genetics
TL;DR: Epidermal DNA synthesis induced by 2 nmol TPA was potentiated by mirex, further supporting a role for mireX in potentiation of epidermal TPA activity. (via Semantic Scholar)
