Effect of calcitonin gene related peptide vs sodium nitroprusside to increase temperature in spontaneous canine tumours during local hyperthermia

INTERNATIONAL JOURNAL OF HYPERTHERMIA, 20(5), 477–489.

By: J. Poulson*, Z. Vujaskovic*, A. Gaskin*, S. Larue*, R. Meyer n, D. Prescott, T. Samulski*, D. Thrall n, M. Dewhirst*

author keywords: tumours; hyperthermia; sodium nitroprusside; CGRP
MeSH headings : Animals; Blood Pressure / drug effects; Calcitonin Gene-Related Peptide / therapeutic use; Combined Modality Therapy; Dog Diseases / drug therapy; Dog Diseases / physiopathology; Dog Diseases / radiotherapy; Dog Diseases / therapy; Dogs; Hyperthermia, Induced; Nitroprusside / therapeutic use; Sarcoma / drug therapy; Sarcoma / radiotherapy; Sarcoma / therapy; Sarcoma / veterinary; Soft Tissue Neoplasms / drug therapy; Soft Tissue Neoplasms / radiotherapy; Soft Tissue Neoplasms / therapy; Soft Tissue Neoplasms / veterinary; Vasodilator Agents / therapeutic use
TL;DR: There is little justification for translation of this approach to human trials using conventional local hyperthermia treatment of SNP, and a significant increase was seen in tumour but not normal subcutaneous tissue temperatures, which was not sufficient to improve the likelihood of increased response rates. 2000 journal article

Local 42 degrees C hyperthermia improves vascular conductance of the R3230Ac rat mammary adenocarcinoma during sodium nitroprusside infusion

RADIATION RESEARCH, 154(2), 196–201.

By: R. Meyer n, R. Braun*, G. Rosner* & M. Dewhirst*

TL;DR: Local 42°C tumor heating, combined with a moderate reduction in blood pressure with nitroprusside, overrides the vascular steal effect associated with reduced perfusion pressure alone and results in improved tumor perfusion. (via Semantic Scholar)

By: Z. Vujaskovic*, J. Poulson*, A. Gaskin*, D. Thrall n, R. Page n, H. Charles*, . MacFall*, D. Brizel* ...

author keywords: tumors; hyperthermia blood how; pO(2); pH; thermal dose
MeSH headings : Animals; Combined Modality Therapy; Dogs; Female; Hydrogen-Ion Concentration; Hyperthermia, Induced / methods; Male; Oxygen / metabolism; Partial Pressure; Sarcoma, Experimental / blood supply; Sarcoma, Experimental / metabolism; Sarcoma, Experimental / radiotherapy; Sarcoma, Experimental / therapy; Soft Tissue Neoplasms / blood supply; Soft Tissue Neoplasms / metabolism; Soft Tissue Neoplasms / radiotherapy; Soft Tissue Neoplasms / therapy
TL;DR: This study suggests that hyperthermia has biphasic effects on tumor physiologic parameters, and suggests that the thermal threshold for vascular damage is higher in spontaneous tumors than in more rapidly growing rodent tumors. (via Semantic Scholar)
