Immobilization and utilization of the recombinant fusion proteins trypsin-streptavidin and streptavidin-transglutaminase for modification of whey protein isolate functionality

JOURNAL OF AGRICULTURAL AND FOOD CHEMISTRY, 50(13), 3723–3730.

By: C. Wilcox n, D. Clare, V. Valentine n & H. Swaisgood n

author keywords: immobilization; trypsin; transglutaminase; whey protein; functionality
MeSH headings : Adsorption; Biotinylation; Chemical Phenomena; Chemistry, Physical; Cross-Linking Reagents; DNA, Recombinant; Dithiothreitol / pharmacology; Enzymes, Immobilized; Escherichia coli / genetics; Gels; Glass; Milk Proteins / chemistry; Milk Proteins / metabolism; Recombinant Fusion Proteins; Streptavidin / genetics; Temperature; Transglutaminases / genetics; Transglutaminases / metabolism; Trypsin / genetics; Trypsin / metabolism; Viscosity; Whey Proteins
TL;DR: By combining limited proteolysis with controlled cross-linking, it was possible to create derivatives of whey proteins with enhanced functional properties that allowed for recycling of the enzyme, eliminated the requirement for a downstream inactivation step, and permitted control over the extent of modification. JOURNAL OF DAIRY SCIENCE, 85(8), 1903–1908.

By: C. Wilcox n, V. Janolino n & H. Swaisgood n

author keywords: CD36; milk fat globule membrane; skim milk; sulfhydryl oxidase
MeSH headings : Amino Acid Sequence; Animals; Blotting, Western; CD36 Antigens; Cattle; Centrifugation; Chromatography, Gel; Electrophoresis, Polyacrylamide Gel; Glycolipids / analysis; Glycoproteins / analysis; Glycosylation; Lipid Droplets; Membrane Glycoproteins / chemistry; Membrane Glycoproteins / isolation & purification; Milk / chemistry; Molecular Sequence Data; Molecular Weight; Organic Anion Transporters / chemistry; Organic Anion Transporters / isolation & purification; Ultrafiltration
TL;DR: CD36, a common milk fat globule membrane glycoprotein, was isolated from skim milk by methods similar to those previously utilized for the isolation of sulfhydryl oxidase, giving similar purity as observed by electrophoresis and suggested tissue-specific glycosylation. (via Semantic Scholar)

JOURNAL OF AGRICULTURAL AND FOOD CHEMISTRY, 50(20), 5546–5551.

By: C. Wilcox n & H. Swaisgood n

author keywords: immobilization; transglutaminase; viscosity; whey protein isolate; gelation; cross linking
MeSH headings : Adsorption; Avidin / chemistry; Bacteria / enzymology; Biotinylation; Cross-Linking Reagents; Enzymes, Immobilized; Glass; Microspheres; Milk Proteins / chemistry; Rheology; Solutions; Transglutaminases / metabolism; Viscosity; Whey Proteins
TL;DR: The functional properties of several batches of WPI were modified using <10 mg of the same enzyme, illustrating the capacity of immobilized enzymes to be used more frequently in applications of this nature. (via Semantic Scholar)
