TL;DR: This work expanded previous synthetic efforts by preparing selected aryl derivatives of the lipoxazolidinones and further evaluating the potential to expand the activity of this class of molecules to Gram-negative pathogens, and utilized a TolC knockout strain of E. coli to demonstrate that these compounds are subject to efflux in Gram- negative pathogens. TL;DR: The first synthesis of lipoxazolidinone A is reported, with 15 structural analogues to explore its active pharmacophore, and initial resistance and mechanism of action studies suggest that 4-oxzolidinones are valuable scaffolds for antimicrobial development and reveal simplified lead compounds for further optimization.
