Identification of a Campylobacter coli methyltransferase targeting adenines at GATC sites

Dutta, V., Altermann, E., Crespo, M. D., Olson, J. W., Siletzky, R. M., & Kathariou, S. (2016, November 19). FEMS Microbiology Letters, Vol. 364.

By: V. Dutta n, E. Altermann*, M. Crespo n, J. Olson n, R. Siletzky n & S. Kathariou n

MeSH headings : Adenine / metabolism; Animals; Campylobacter coli / enzymology; Campylobacter coli / genetics; Cattle; DNA Methylation; Genome, Bacterial; Humans; Methyltransferases / genetics; Methyltransferases / isolation & purification; Methyltransferases / metabolism; Nucleotide Motifs; Open Reading Frames; Sequence Analysis / methods
topics (OpenAlex): Salmonella and Campylobacter epidemiology; Enterobacteriaceae and Cronobacter Research; Vibrio bacteria research studies
TL;DR: To determine whether ORF0059 mediated MboI resistance and hence encoded a putative N6‐adenine DNA methyltransferase, the gene was cloned immediately upstream of a chloramphenicol resistance cassette (cat) and a PCR fragment harboring ORF 0059‐cat was transformed into C. coli 11601, suggesting a direct role of this gene in methylation of adenines at GATC sites. Crespo, M. D., Altermann, E., Olson, J., Miller, W. G., Chandrashekhar, K., & Kathariou, S. (2016, June 5). Plasmid, Vol. 86, pp. 32–37.

author keywords: Campylobacter; C. jejuni; Plasmid; Antibiotic resistance; tet(O); Tetracycline resistance; Kanamycin resistance; aphA-3
MeSH headings : Animals; Anti-Bacterial Agents / pharmacology; Bacterial Proteins / genetics; Base Composition; Base Sequence; Campylobacter jejuni / drug effects; Campylobacter jejuni / genetics; Campylobacter jejuni / isolation & purification; Carrier Proteins / genetics; Conjugation, Genetic / genetics; DNA, Bacterial / genetics; Jejunum / microbiology; Kanamycin / pharmacology; Kanamycin Kinase / genetics; Microbial Sensitivity Tests; North Carolina; Plasmids / genetics; Poultry Diseases / microbiology; Sequence Analysis, DNA; Tetracycline / pharmacology; Tetracycline Resistance / genetics; Turkeys / microbiology
topics (OpenAlex): Salmonella and Campylobacter epidemiology; Viral gastroenteritis research and epidemiology; Bacteriophages and microbial interactions
TL;DR: A novel plasmid harboring tet(O) was identified in C. jejuni strain 11601MD, which was isolated from the jejunum of a turkey produced conventionally in North Carolina, and will contribute to a better understanding of the distribution of potentially self-mobilizing plasmids harboring antibiotic resistance determinants in Campylobacter spp. (via Semantic Scholar)

Routes of transmission of Salmonella and Campylobacter in breeder turkeys

Crespo, M. D., Kathariou, S., Grimes, J. L., Cox, N. A., Buhr, R. J., Frye, J. G., … Smith, D. P. (2016, August 6). The Journal of Applied Poultry Research.

author keywords: Salmonella; Campylobacter; breeder turkeys; routes of transmission
topics (OpenAlex): Salmonella and Campylobacter epidemiology; Animal Nutrition and Physiology; Viral gastroenteritis research and epidemiology
TL;DR: Monitoring of turkey breeder hens and toms from d of hatch to 65 wk of age showed that Campylobacter spread rapidly and cross-contaminated turkeys throughout the growout house, confirming the importance of proper pest control and biosecurity to reduce the spread of the bacteria. By: M. Crespo n, J. Olson n, E. Altermann n, R. Siletzky n & S. Kathariou n

MeSH headings : Animals; Campylobacter Infections / microbiology; Campylobacter Infections / veterinary; Campylobacter coli / drug effects; Campylobacter coli / genetics; Campylobacter coli / isolation & purification; Carrier Proteins / genetics; Chromosomes, Bacterial; DNA Transposable Elements; DNA, Bacterial / chemistry; DNA, Bacterial / genetics; Genes, Bacterial; Molecular Sequence Data; Mutagenesis, Insertional; Poultry Diseases / microbiology; Sequence Analysis, DNA; Swine; Swine Diseases / microbiology; Tetracycline Resistance; Turkeys
topics (OpenAlex): Salmonella and Campylobacter epidemiology; Viral gastroenteritis research and epidemiology; Plant Pathogenic Bacteria Studies
TL;DR: In turkey-derived Campylobacter coli isolates of a unique lineage (cluster II), the tetracycline resistance determinant tet(O) was chromosomal and was part of a gene cassette (transposon) interrupting a Campyloblacter jejuni-associated putative citrate transporter gene.
