Mucosal Immunogenicity of Genetically Modified Lactobacillus acidophilus Expressing an HIV-1 Epitope within the Surface Layer Protein

PLOS ONE, 10(10).

MeSH headings : Adaptive Immunity; Animals; Antibodies, Bacterial / immunology; Antibody Formation / immunology; Antibody Specificity / immunology; Bacterial Proteins / genetics; Bacterial Proteins / immunology; Cytokines / metabolism; Epitopes / genetics; Epitopes / immunology; HIV-1 / genetics; HIV-1 / immunology; Humans; Immunity, Mucosal; Immunization; Immunoglobulin A / immunology; Lactobacillus acidophilus / genetics; Lactobacillus acidophilus / immunology; Mice; Models, Animal; Mucous Membrane / immunology; Mucous Membrane / microbiology; Mucous Membrane / virology; Recombinant Fusion Proteins / genetics; Recombinant Fusion Proteins / immunology
TL;DR: The constructed genetically modified Lactobacillus acidophilus strains expressing the membrane proximal external region (MPER) from human immunodeficiency virus type 1 (HIV-1) within the context of the major S-layer protein, SlpA, demonstrated the potential use of the LactOBacillus S- layer protein for development of oral vaccines targeting specific peptides. By: A. Kajikawa*, L. Zhang*, J. Long n, S. Nordone n, L. Stoeker n, A. LaVoy*, S. Bumgardner n, T. Klaenhammer n, G. Dean*

MeSH headings : AIDS Vaccines / administration & dosage; AIDS Vaccines / genetics; AIDS Vaccines / immunology; Administration, Oral; Animals; Bacterial Vaccines / administration & dosage; Bacterial Vaccines / genetics; Bacterial Vaccines / immunology; Cell Surface Display Techniques / methods; Cells, Cultured; Dendritic Cells / immunology; Female; Flagellin / genetics; Flagellin / immunology; HIV-1 / genetics; HIV-1 / immunology; Humans; Intestinal Mucosa / immunology; Lactobacillus acidophilus / genetics; Leukocytes, Mononuclear / immunology; Mice; Mice, Inbred BALB C; Salmonella typhimurium / genetics; Salmonella typhimurium / immunology; Vaccines, Synthetic / administration & dosage; Vaccines, Synthetic / genetics; Vaccines, Synthetic / immunology; gag Gene Products, Human Immunodeficiency Virus / genetics; gag Gene Products, Human Immunodeficiency Virus / immunology
TL;DR: Results suggested that the Gag-displaying L. acidophilus elicited specific immune responses and the coexistence of FliC conferred an adjuvant effect on local IgA production. (via Semantic Scholar)
