Using an intergenerational pig model of diet restriction (DR) that produces reduced weight gain, we developed a feature-selection algorithm to identify representative characteristics distinguishing DR fecal microbiomes from those of full-fed (FF) pigs as both groups consumed a common sequence of diets during their growth cycle. Gnotobiotic mice were then colonized with DR and FF microbiomes and subjected to controlled feeding with a pig diet. DR microbiomes have reduced representation of genes that degrade dominant components of late growth-phase diets, exhibit reduced production of butyrate, a key host-accessible energy source, and are causally linked to reduced hepatic fatty acid metabolism (β-oxidation) and the selection of alternative energy substrates. Methods A total of 27 pregnant sows were randomly assigned into three treatment groups. Each group was fed a standard diet (3265 kcal ME/kg) supplemented with either 0, 0.25% or 0.5% clofibrate (w/w) from d 107 of gestation to d 7 of lactation. A piglet from each litter was sampled on 1, 7, 14 and 19 d of age. Fatty acid oxidation was examined in homogenates of ileum mucosa using 1 mM [1–14C] oleic acid as a substrate in the absence or presence of in vitro supplemented L-carnitine (1 mM), malonate (10 mM) or carnitine + malonate. Results The metabolism of [1–14C] oleic acid was modified by the maternal clofibrate, and the effects varied with the dose of clofibrate and postnatal age (p < 0.001). The 14C accumulation in 14CO2 increased by 2.4 fold at d7 (vs d1), but showed no difference after d7. Maternal clofibrate stimulated the 14CO2 accumulation from d1 to d19, but the stimulation decreased with postnatal age. The 14C accumulation in acid soluble products (14C-ASP) was reduced by 8.4 fold at d7 (vs d1), and no difference was observed after d7. Maternal clofibrate increased 14C-ASP at d1, and the effect was greater as maternal clofibrate was incremented from 0.25 to 0.5%. The 14C accumulation in esterified products (14C-ESP) was greater at d7 than d1, 14 and 19. Maternal clofibrate reduced the 14C-ESP and, again the reduction was stronger with increasing clofibrate dose. In vitro supplementation of carnitine increased 14CO2 by 60% and 14C-ASP by 120%, but reduced 14C-ESP by 39% compared to control incubations. Supplementation of malonate reduced 14CO2 by 95% and 14C-ESP by 44%, but no effect on 14C-ASP was detected. Conclusions Maternal dietary supplementation of clofibrate increases intestinal fatty acid oxidation in suckling offspring, but the increase is attenuated after one week. Inhibition of the TCA cycle reduces CO2 production without affecting ASP. The availability of carnitine could be a key element to maintain a high rate of intestinal fatty acid oxidation during the neonatal period. However, the effects of maternal undernutrition have generally focused on the reduced maternal nutrient supply to the fetus. The potential impairment of fetal metabolic pathways has not been well studied. Methods Pregnant gilts (Landrace x Yorkshire x Duroc) received the NRC gestation diet with (n = 4) or without (n = 4) 50% intake restriction at insemination day and 70% for the following gestation period. Full term fetuses were obtained via C-section, two piglets were selected from each gilt in both groups and subject to hepatic tissue collection. MicroRNA and mRNA deep sequencing analysis was performed using the Illumina GAIIx system. The mRNA-miRNA correlation and associated signaling pathways were analyzed via CLC workbench, Ingenuity Pathway Analysis Software. Results A total of 42 differentially expressed miRNAs were identified between intake-restriction and full-nutrition group. Among of these, mir-206, mir-133b, mir-1246, mir-1843 and mir-7139 are the most downregulated and mir-10b, mir-708 and mir-222 are the most upregulated miRNAs. A total of 1215 mRNAs were identified to differentially expressed between two groups. Two metabolic pathways: retinol biosynthesis and oxidative phosphorylation were significantly modified, and the modification was associated with the miRNA changes induced by the maternal feed restriction. Briefly, the retinol biosynthesis pathway was upregulated (p < 0.01), in which those differential expressed mir-221, mir-4492, mir-1281 and mir-4492 were predicted targeting genes AADAC, CES3, PNPLA3 and RDH13 in the pathway. The oxidative phosphorylation pathway was upregulated (p < 0.05), and those differential expressed mir-1843, mir-222 and mir-184 were predicted targeting genes ATP5F1C, NDUFA1, NDUFB10, and NDUFS7 in this pathway. Conclusions These results provide the framework for further understanding of negative impact of maternal malnutrition on hepatic metabolic pathways via miRNA-RNA interactions in full-term fetal pigs. Suckling newborn pigs (n = 72) were assigned into 8 dietary treatments following a 2 × 4 factorial design: ± clofibrate (0.35%) and diets containing 5% of either (1) glycerol-succinate (GlySuc), (2) tri-valerate (TriC5), (3) tri-hexanoate (TriC6), or (4) tri-2-methylpentanoate (Tri2MPA). Pigs were housed individually and fed the iso-caloric milk replacer diets for 5 d. Renal fatty acid oxidation was measured in vitro in fresh tissue homogenates using [1-14C]-labeled palmitic acid. The oxidation was 30% greater in pig received clofibrate and 25% greater (p < 0.05) in pigs fed the TriC6 diet compared to those fed diets with GlySuc, TriC5, and Tri2MPA. Addition of carnitine also stimulated the oxidation by twofold (p < 0.05). The effects of TriC6 and carnitine on palmitic acid oxidation were not altered by clofibrate stimulation. However, renal fatty acid composition was altered by clofibrate and Tri2MPA. Its action, metabolism, half-life and excretion have been well documented in liver of adult humans and rodent species, but it has not been evaluated well in extrahepatic tissues in neonates as a stimulator of energy utilization. In this study, the role of maternal administration of clofibrate in development of renal fatty acid oxidation was evaluated using swine as a model. Methods A randomized complete block design was used with a total of 27 pregnant sows. The sows were fed standard gestation-lactation diets supplemented with either 0 (control), 0.25% or 0.5% clofibrate (w/w) from d 107 of gestation to d 7 of lactation. Fatty acid oxidation was measured in the presence or absence of carnitine (1 mM) or/and malonate (5 mM) in fresh kidney homogenates from piglets at d1, 7, 14 and 19 of age using 14C-oleic acid (1 mM) as substrate (9.9 mBq/mmol). Results Interactions (P < 0.001) were observed between maternal clofibrate levels and postnatal age on 14C accumulation in CO2 (14CO2), acid soluble products (14C-ASP) and esterified products (14C-ESP). The 14CO2 increased by 1.3 fold from d1 to d7, but showed no differences between d7, 14 and 19 in pigs from the control sows. Maternal supplementation of clofibrate increased 14CO2 in pigs across all ages, but the increase was higher in pigs from sows fed 0.5% versus 0.25% clofibrate at d14. The 14C-ASP was 7-fold higher in d1 pigs from control sows than all other ages. Maternal supplementation of clofibrate increased 14C-ASP by 2 fold in pigs at d1, but had no detectable effect at d 7, 14 and 19. The 14C-ESP increased from d1 to d7 and decreased from d7 to d19 in pigs from control sows. Maternal supplementation of clofibrate had no detectable impact on 14C-ESP at d1, 14 and 19, but decreased 14C-ESP measured in d7 pigs. In vitro carnitine supplementation increased 14CO2 and malonate supplementation decreased 14CO2, but neither carnitine nor malonate altered 14C-ASP or 14C-ESP. Fresh tissue homogenates were assayed under nine concentrations of γ-BB from 0 to 1.5 mM. Substrate inhibition associated with age was observed at ≥0.6 mM of γ-BB. Hepatic activity was low at birth but increased after 1 day. By 21 days, the activity rose by 6.6-fold ( P < 0.05) and remained constant after 56 days. This study was to determine if 30% extra supplemental dl-methionine alleviated or prevented those adverse effects of a higher stocking density in broiler chickens. A total of 560 male Cornish Cross cockerels (day old) were divided into four groups: two stocking densities (9 and 12 birds/m2) and two supplementations of methionine (grower: 2.90 or 3.77 g/kg and finisher: 2.60 or 3.38 g/kg). Growth performance was recorded weekly. Blood and tissues were sampled at the end of each period. High stocking density decreased (P < 0.05) body weight and growth performance of growers and (or) finishers. Those differences were partially attenuated by the extra methionine supplementation. The high methionine elevated (P < 0.05) glutathione (GSH) concentration in the thigh at both ages (> 24%). The high stocking density elevated (>28%, P < 0.05) glutathione concentration in the plasma, breast, and thigh of growers, but decreased (P < 0.05) it in the liver of growers and thigh of finishers. Interaction effects (P < 0.05) between dietary methionine and stocking density were found on activities of the antioxidant enzyme glutathione S-transferase in the liver of growers and breast, thigh, and adipose tissue of finishers. The interaction effect was also found on activities of glutathione peroxidase and superoxide dismutase in the thigh of growers. Chicks were fed the same corn-soybean meal-based starter diet and then experimental diets (grower: 6.4 g SAA/kg, finisher: 5.9 g SAA/kg, n = 10 pens/treatment). Growth performance and mortality were recorded, and blood and tissue samples (3 chicks/pen) were collected at the end of each period. Data were analyzed by two-way ANOVA. High stocking density decreased (P < 0.05) BW and (or) ADG of growers and finishers. Both high stocking density and methionine decreased (P < 0.05) ADFI and the high methionine improved (P < 0.05) feed use efficiency of chicks during the grower-finisher period. Neither treatment affected feather coverage, mortality, incidences of diseases, or dressing percentage. The high methionine improved (P < 0.05) meat to bone ratio of thigh, and exerted mixed effects (P < 0.05) on texture of breast and thigh. Dietary methionine and stocking density showed no major effects on tissue lipid profiles, but moderate effects (P < 0.05) on plasma concentrations of total cholesterol and(or) triglycerides of chicks. Fatty acid profiles in the liver and breast of chicks were unaffected by either treatment. The two treatments enhanced (P < 0.05) tibial maximum breaking strength (15–20%) and plasma alkaline phosphatase activity of growers. In conclusion, the high stocking density affected growth performance, meat quality, and health status of broilers. Supplementing 30% extra dietary methionine seemed to mitigate some of those effects. The Edu assays were performed to analyze cell proliferation. To analyze cell differentiation, the expressions of myogenic marker genes were determined at the transcriptional and translational levels by qRT-PCR, immunoblotting and immunofluorescence. Global gene expression patterns were characterized using RNA-sequencing. Phosphorylation levels of ERK and Akt were examined by immunoblotting. Cell viability and proliferation was significantly inhibited after incubation with EPA (50 and 100 μM) or DHA (100 μM). Both EPA and DHA suppressed C2C12 myoblasts differentiation. RNA-sequencing analysis revealed that some muscle-related genes were significantly downregulated following EPA or DHA (50 μM) treatment, including insulin-like growth factor 2 (IGF-2), troponin T3 (Tnnt3), myoglobin (Mb), myosin light chain phosphorylatable fast skeletal muscle (Mylpf) and myosin heavy polypeptide 3 (Myh3). IGF-2 was crucial for the growth and differentiation of skeletal muscle and could activate the PI3K/Akt and the MAPK/ERK cascade. We found that EPA and DHA (50 μM) decreased the phosphorylation levels of ERK1/2 and Akt in C2C12 myoblasts. Pigs (n = 64; 6.53 ± 0.12 kg BW) weaned at 21 d of age were blocked by litter and BW and randomly assigned within blocks to 1 of 2 dietary treatments in a 35-d study. Four pigs were housed per pen, using a total of 16 pens (8 replicate pens per treatment). Treatments were: 1) control diet (without growth-promoting minerals or antibiotics) without PP, and 2) control diet with 4% of PP replacing SBM on a 1:1 basis. Diets were fed in 3 feeding phases (7, 14, and 14 days, respectively) and PP was included in all phases. Supplementation of PP tended (P = 0.07) to increase ADG (204 vs. 150 g/d) and improved (P = 0.02) gain:feed (812 vs 572 g/kg) during wk 2 only. Individual diarrhea scores (scale from 0=very hard to 6=watery diarrhea) determined using fecal loops changed over time (P < 0.001; 2.1, 2.4, 4.1 and 3.5 for d 1, 3, 7, and 14 respectively), but were not affected by PP. No differences were detected for intestinal morphology measured on d 35 in duodenum, jejunum and ileum. Cell proliferation using Ki-67 staining was increased by PP (86.6 vs. 72.2 positive cells) only in the ileum (P = 0.03). No differences in mucosal concentrations of IL-10, TNF-α, and IFN-γ in the duodenum, jejunum or ileum were detected. Number of eosinophils in blood decreased (14.1 vs. 34.8 cells/µL; P = 0.004), while red blood cell counts (5.9 vs 6.2 x 106 cells/µL; P = 0.09) and neutrophil counts (529 vs. 695 cells/µL; P = 0.07) tended to decrease with PP. We hypothesized that Asn can mitigate LPS-induced injury of intestinal structure and barrier function by regulating inflammatory signaling pathways. We executed the following experiment using weanling pigs for each of the groups: (1) non-challenged control; (2) LPS-challenged control; (3) LPS + 0.5% Asn; (4) LPS + 1.0% Asn. After 21-d feeding, pigs received an i.p. injection of either saline or LPS. Four h after injection, the mid-jejunum and mid-ileum samples were collected. We found that Asn restored ODC expression that was decreased by LPS treatment. Asn also restored intestinal morphology and barrier function that were impaired by LPS treatment. In addition, Asn down-regulated intestinal caspase-3 protein expression and TNF-α concentration, and decreased the mRNA expression of intestinal TLR4, TLR4 downstream signals (myeloid differentiation factor 88, IL-1 receptor-associated kinase 1 and TNF-α receptor-associated factor 6 and NOD1, NOD2 and their adaptor molecule (receptor-interacting serine/threonine-protein kinase 2). Moreover, Asn decreased p38 phosphorylation but increased ERK1/2 phosphorylation. Our results suggest that Asn improves intestinal integrity during an inflammatory insult, which appears to be related to the decrease of intestinal pro-inflammatory cytokine (via TLR4, NODs and p38) Its precocial development at birth affords ready adaptation to artificial rearing systems, and research using this model spans a wide array of nutrients. Sophisticated in vitro and in vivo methodologies supporting both invasive, reduction-science research as well as whole-animal preclinical investigations have been developed. Potential applications may dually benefit both agricultural and medical sciences (e.g., “agrimedical research”). The broad scope of this review is to outline the fundamental elements of the piglet model and to highlight key aspects of relevance to various macronutrients, including lipids, carbohydrates, proteins/amino acids, and calcium/phosphorus. The birth order and weight of 745 piglets (from 75 litters) were recorded during a one-week period of farrowing. Only pigs weighing greater than 0.68 kg birth weight were chosen for the trial. Sow colostrum was collected during parturition, and piglets were bled between 48 and 72 hours post-birth. Piglet serum IgG and colostral IgG concentrations were determined by radial immunodiffusion. Results Sow parity had a significant (P < 0.001) effect on sow colostral IgG concentration, being 5% higher in multiparous females. Sow colostral IgG concentration explained 6% and piglet birth order accounted for another 4% of the variation observed in piglet serum IgG concentration (P < 0.05); however, birth weight had no detectable effect. Piglet serum IgG concentration had both a linear (P < 0.05) and quadratic effect (P < 0.05) on % survival. Piglets with 1,000 mg/dl serum IgG or less (n=24) had a 67% survival; whereas, piglets with IgG concentrations between 2250 to 2500 mg/dl (n=247) had a 91% survival. Birth order had no detectable effect on survival, but birth weight had a positive linear effect (P < 0.05). Piglets weighing 0.9 kg (n = 107) at birth had a 68% survival rate, and those weighing 1.6 kg (n = 158) had an 89% survival. Conclusion We found that the combination of sow colostrum IgG concentration and birth order can account for 10% of the variation of piglet serum IgG concentration and that piglets with less than 1,000 mg/dl IgG serum concentration and weight of 0.9 kg at birth had low survival rate when compared to their larger siblings. Studies using piglets have illustrated the divergent nature of intermediary metabolism in this model, wherein ketogenesis and beta-oxidation are small and acetogenesis is an important route of fuel carbon flux. To clarify potential species differences in hepatic lipid metabolism and its control, we compared O2 consumption and metabolic end products in fasted pig and rat liver homogenates treated with 1-[14C]C16:0. Carboxyl carbon accumulation in acid-soluble products (ASP) plus CO2 was threefold greater and O2 consumption was twofold greater in rats (P < 0.05). Unlike rats, pigs showed negligible carboxyl carbon accumulation in ketone bodies (3-7% of ASP), whereas acetate was a carboxyl carbon reservoir in both animals (17-31% of ASP in pigs). Malonate increased (approximately 2-fold) and antimycin/rotenone decreased (40-60%) radiolabel accumulation in acetate. The differences between odd and even chain-length and the impacts of valproate and malonate upon the rate of β-oxidation and ASP characteristics were tested. A minor amount of fatty acid carboxyl carbon (⩽ 10% of organic acids identified by radio-HPLC) accumulated in ketone bodies regardless of chain-length or inhibitor used. In all cases, acetate represented the major reservoir of carboxyl carbon, accounting for 60–70% of radiolabel in identified organic acids. Cells given [1-14C]C7:0 accumulated 85% more carboxyl carbon in Krebs cycle intermediates when compared with C8:0, while accumulation in acetate was unaffected.
