AU - Dietze, Reynaldo T2 - The American Journal of Tropical Medicine and Hygiene AB - Although Brazilian spotted fever is known to occur in several adjacent states, infection with spotted fever group (SFG) rickettsiae has not previously been documented in the Brazilian state of Espirito Santo. We report a cluster of two proven and four suspected cases of Brazilian spotted fever in a small valley near Colatina, Espirito Santo. Four patients died without confirmatory serologic or pathologic studies. The two surviving patients had serologic and/or pathologic evidence of infection with SFG rickettsiae. Results of a survey done in 1991 confirmed the presence of antibodies reactive with SFG rickettsiae in the serum of dogs living in this region. Two of 25 dogs had elevated microimmunofluorescent antibody titers (1:64 and 1:256). Both seropositive dogs were pets of the index cases and lived in the valley where all six human cases were presumed to have acquired their infections. These cases and the results of the canine serosurvey suggest that a focus of virulent disease exists in an area not previously known to be endemic for SFG rickettsiae. An area of focal intense uptake was identified in the area of the right adrenal gland. A pheochromocytoma was confirmed histologically after surgical excision. T2 - Journal of Small Animal Practice AB - ABSTRACT This paper describes the clinical and pathological features of 11 dogs with insulin‐secreting tumours of the pancreas. All the dogs showed episodic weakness or collapse. The diagnosis was made on fasting plasma glucose and serum insulin concentrations, the insulimglucose ratio, and the results of an intravenous glucose tolerance test. Ten of the dogs had exploratory laparotomy, and partial pancreatectomy was performed in nine of the cases. One case was euthanased at surgery because of widespread metastases. The tumours were graded histologically and the results compared with the time to recurrence of clinical signs and postoperative survival time. Postoperative survival time for dogs which died or were euthanased as a direct result of tumour recurrence, and time to recurrence of clinical signs were calculated from actuarial survival curves. The median time to recurrence of clinical signs after surgery was 12 months (range from four to 16 months; mean time to recurrence of clinical signs 12 months). Two cases died of unrelated disease, without recurrence of hypoglycaemic signs. The median postoperative survival time was 14 months (range 10 to 33 months; mean survival time 15 months). There is a suggestion that tumours with a high mitotic count carried a worse prognosis. T2 - Journal of Veterinary Internal Medicine AB - The prevalence of feline thrombocytopenia (< 200,000 platelets/microL) at North Carolina State University, College of Veterinary Medicine Teaching Hospital, from January 1985 to March 1990, was 1.2% (41/3300). Cats were divided into six categories based on clinical diagnoses: 29% (12/41) had infectious disease, 20% (8/41) had neoplasia, 7% (3/41) had cardiac disease, 2% (1/41) had primary immune-mediated disease, 22% (9/41) had multiple diseases, and 20% (8/41) had disorders of unknown etiology. The mean platelet count for all thrombocytopenic cats was 52,000/microL +/- 46,000/microL (1 SD) with a range of 1000-190,000/microL. No significant differences were found between groups with respect to platelet count, packed cell volume, or white blood cell count, though anemia and leukopenia were common among the cats as a whole. Bleeding disorders (hemorrhage or thrombosis) were observed in 29% (12/41) of thrombocytopenic cats and were more likely to be associated with neoplasia, cardiac disease, and platelet counts less than or equal to 30,000/microL. Disseminated intravascular coagulopathy was diagnosed in 12% (5/41) of the cats. Infections and/or neoplasia affecting the bone marrow were the most common diseases associated with thrombocytopenia. Feline leukemia virus and myeloproliferative neoplasia accounted for approximately 44% (18/41) of the specific diagnoses in thrombocytopenic cats. Protein values ranged from 75.9% to 97.3% of dry matter. Fat ranged from 0.6% to 9.1% of dry matter. A protein corresponding to beta-lactoglobulin on gel filtration chromatography was the predominant whey protein. Neutralizing antibodies to nine caliciviruses were detected in one sample. Composition of these two samples differs from previous reports for cetacean milk, perhaps due to the stage of lactation. T2 - Journal of Small Animal Practice AB - ABSTRACT The orientation of an ultrasound transducer required to provide standard echocardiographic views of the heart valves that would permit optimal alignment with blood flow for pulsed‐wave Doppler studies, was investigated in 20 normal dogs. Orientation was defined by the site on the thoracic wall at which the transducer was placed and by the angulation and rotation required to produce a two‐dimensional image in which the pulsed‐wave Doppler beam could be aligned with flow through each of the four heart valves. Two views, each a relatively short‐axis projection at the heart base, from either side of the thorax, gave alignment with the pulmonary artery. Three views gave reliable alignment with the aortic valve, from the cardiac apex on each side of the thorax and subcostally. One view was available for the mitral and tricuspid valves, from the left side of the thorax. Definition of these sites should help clinicians and technicians to develop a systematic routine for Doppler investigations in dogs and facilitate communication between investigators. The Nd:YAG laser was applied to six endometrial sites, in each of five uterine specimens, with power densities of 5659 to 33,954 J/cm 2 . Depth of tissue ablation was measured and graded on histologic sections of the tissue lesions. Power density had a significant effect on the depth of tissue ablation (p < .001). Grade 3 lesions (full‐thickness ablation of the endometrium) were created with energy densities of 16,977 to 33,954 J/cm 2 . Six mares had endometrial cysts treated by photoablation. Two of the four mares that were reproductively sound but barren, despite appropriate breeding, produced foals after treatment. One mare remained reproductively unsound after treatment, and another mare that was treated postpartum was bred successfully. Lesions noted were paronychia, ulcers on the palate, oropharynx, gums and tongue, as well as ulceration and crusts on the metacarpal, metatarsal and digital pads. Histology of lesions showed dermal‐epidermaj separation and ulceration. Immunohistochemistry showed collagen IV (characteristic of the lamina densa) located at the roof of the cleft at both the dermal‐epidermal and follicular‐dermal interfaces. Electron microscopy showed the dermal‐epidermal cleft to be sublamina densa. Résumé— Une épidermolyse bulleuse dystrophíque a été diagnostiquée chez un chat européen à poils courts de huit mois. Les lésions étaient une paronychie, des ulcères du palai, de l'oropharynx, des gencives et de la langue ains que des ulcères et des croutes au niveau des métacarpes, métatarses et coussinets plantaires. L'examen histologique des lésions faiasait apparaitre des ulcérations et des disjonctions derme/épiderme. L'étude immunohistochimique a montré la présence de fibres de collagène IV (caractéristiques de la lamina densa) sur lew toit des bulles tant au niveau des interfaces derme/épiderme que derme/follicule. La microscopie électronique a montré que les séparations derme/épiderme se faisaient au niveau de la sublamina densa. Zusammenfassung— Eine dystrophe Epidermolysis bullosa wurde bei einer achtjährigen kurzhaarigen männlichen hauskatze diagnostiziert. Die beschriebenen Veränderungen bestanden in Paronychie, Ulzera an Gaumen, Oropharynx, Gingiva und Zunge sowie Ulzerationen und Krusten an den metacarpalen, metatarsalen und digitalen Ballen. Die Histologie der Veränderungen zeigte dermal‐epidermale Trennungen und Ulzerationen. Mit der Immunhistochemie wurde Kollagen IV nachgewiesen (ein Kennzeichen der Lamina densa), das am Gewölbe der Spaltbildung sowohl an den derma‐epidermalen als auch an den follikulär‐dermalen Schnittstellen lokalisiert war. Die Elektronenmikroskopie zeigte, daß die dermal‐epidermale Spaltbildung die Sublamina densa darstellte. Resumen En un gato macho de ocho meses doméstico de pelo corto se diagnóstico epidermolisis distrófica bullosa. Las lesiones presentes fueron paroniquia, úlceras del paladar, orofaringe, encías y lengua, así como ulceraciones y costras en las regiones del metacarpo, metatarso, y almohadillas plantares. Las lesiones histologicas consistieron de separación y ulceración a nivel de la dermis y epidermis. Exámenes inmunohistoquímicos demostraron la presencia de colágeno IV (característico de la lámina densa), localizado en el techo de las indentactiones en ambas interfases, epidermo‐dérmica y foliculo‐dérmica. Por medio de microscopía electrónica se comprobo que las indentaciones de la dermis y epidermis consistían de sublámina densa. T2 - Equine Veterinary Journal AB - Summary In an investigation of 2 closely related Miniature Horses with a history of excessive sleepiness, depression and episodes of collapse, a diagnosis of narcolepsy was made on the basis of neurological examination and pharmacological testing. Further investigations included electroencephalographic examination (KEG), and analysis of protein content, cell count and monoamine metabolite concentrations of lumbosacral cerebrospinal fluid (CSF). There were no abnormalities noted in the EKGs, and no consistent changes in CSF neurotransmitter metabolites in the narcoleptic horses when compared with 3 normal, unrelated Miniature Horses and 2 related, clinically unaffected animals. The breeding background of the 2 affected horses was investigated and a limited survey of Miniature Horse breeders in North America was conducted. These investigations have shown that narcolepsy is a rare but distinct syndrome in the Miniature Horse, and that the cases described here appear to represent a familial occurrence of the disease. On the basis of tissue distribution and biochemical characteristics, this antigen is equine MHC II. The equine MHC II antigen was found on a large subset of T-lymphocytes in addition to all B-lymphocytes, as has been reported previously. In addition MHC II was found to be present on a large proportion of both the mutually exclusive equine T-lymphocyte subpopulations which express either the equine homologues of CD4, or CD8. In a study of changes in equine MHC II expression with age it was found that far fewer lymphocytes express MHC II in neonatal foals than in adult horses. An increase with age was demonstrated. This age related change in MHC II expression was shown to occur principally within the T-lymphocyte subpopulation. It is proposed that this change may be associated with the development of a memory T-lymphocyte population, and that MHC II expression may be a marker for memory T-lymphocytes. Rats were anaesthetized with a xylazine/ketamine mixture, heated on a water-bed for 1 h to an intraperitoneal temperature of 41 1 ±0.2°C, and maintained at hyperthermia for an additional 30 min (HY plateau). CDDP (5 mg/kg body weight) was administered at the start of the heating or at the HY plateau. Neither HY alone nor CDDP (5 mg/kg) at normothermia produced a significant effect on weight loss or nephrotoxicity. CDDP administered at the start of heating produced a moderate 1–5–fold increase in serum urea nitrogen (SUN) and serum creatinine concentrations. CDDP administered at the HY plateau produced a significant six-fold increase in SUN and a four-fold increase in creatinine concentration. Weight loss increased two- to three-fold from the combined regimen, but only the rats given CDDP at the HY plateau continued to lose weight through day 7. A loss of kidney copper (5040%) resulted from the combined regimen, similar to losses observed with higher doses of CDDP at normothermia. HY alone had little effect on concentrations of kidney copper or zinc up to 4 days post-treatment. Biomechanical testing, in a controlled environment, was performed during the 28‐day study period. Polydioxanone and chromic gut retained greater than 90% of their original strengths after 28 days of incubation in sterile urine and 87% of original strengths in E. co//‐inoculated urine. Polyglyconate retained 24% and 18% of original strength, respectively, after incubation in sterile and E. co/i‐inoculated urine for 28 days. Polyglycolic acid and PG‐910 retained less than 30% of original strength in sterile urine and only 7% in E. co/i‐inoculated urine after 21 days of incubation. In P. m/rabi/is‐inoculated urine, loss of tensile strength and elongation was significant for all suture materials. Polyglycolic acid and PG‐910 lost all strength after 24 hours of incubation. Polydioxanone lost all strength after 7 days of incubation, whereas GTMC retained 19% at day 7. Chromic gut retained 78% at day 7 and 16% after 21 days of incubation, however, the absence of normal phagocytic destruction of chromic gut in this in vitro study may have artificially elevated these values. In sterile urine with chemically modified pH, loss of strength and elongation was greater in alkaline urine than in neutral or acidic urine for all types of suture materials. The method involved an ultrafiltration of diluted serum/milk with an equal volume of 50% acetonitrile through a 10,000 dalton molecular mass cut-off filter. Separation of ceftiofur and desfuroylceftiofur from the other serum/milk components was performed by ion-paired (octane and dodecanesulfonate) liquid chromatography using a reversed-phase column eluted with acetonitrile-water solution. The ultraviolet-visible absorbance of the column effluent was monitored in 200-350 nm range of a photodiode-array detector or at lambda max 289.6 nm for ceftiofur, lambda max 265.8 nm for desfuroylceftiofur and lambda max 271.4 nm for dimer of desfuroylceftiofur. Recoveries of ceftiofur from bovine milk spiked with 1 and 10 micrograms/ml were 95.9 and 97.0% with coefficients of variation of 3.69 and 2.51%, respectively. Recovery of ceftiofur from bovine serum spiked with 10 micrograms/ml was 90.4% with a coefficient of variation of 5.29%. A correlation coefficient of 0.9992 occurred with ceftiofur in aqueous solutions (n = 5, in duplicates). The limit of detection was estimated to be approximately 50 ppb (ng/ml). Additionally, this paper documents the presence of a ceftiofur metabolite in bovine serum under in vitro and in vivo conditions. The metabolite was identified as desfuroylceftiofur together with its dimer 3,3'-desfuroylceftiofur disulfide by thermospray liquid chromatography-mass spectrometry. Right carotid arteries were surgically elevated to a subcutaneous position for percutaneous catheterization. Each horse was fitted with a device designed to obtain multiple arterial samples while the horse was exercised over a 1.6‐km course. After each horse completed 10 test gallops, the laryngeal hemiplegia was treated using a laryngeal prothesis and ventriculectomy. The horses were then reconditioned, and the exercise test and sampling were repeated. Horses with laryngeal hemiplegia became acidotic, hypoxic, and hypercapnic compared to normal horses. Surgical treatment improved blood gas and acid‐base status, but the values were not equivalent to those in normal horses similarly tested. Twenty-eight dogs were considered evaluable for toxicity. The maximally tolerated dose of CBDCA was conceptually defined as that dose, determined by logistic regression analyses of toxicity data, resulting in a 50% incidence of moderate toxicity (MOD50) or a 5% incidence of severe toxicity (SEV5). Each designated maximally tolerated dose was calculated for the first course of treatment only and for the first and second courses of treatment combined to estimate cumulative drug toxicity. The MOD50 and SEV5 for the first treatment course were 340 and 278 mg/M2, respectively. MOD50 and SEV5 values for the first plus second treatment courses were 327 and 231 mg/M2, respectively. The nadir of neutrophil and platelet counts occurred approximately 14 days after treatment. The mean neutrophil and platelet values for all dogs experiencing myelosuppression during the first two treatment courses were 1541/microL and 62,600/microL, respectively. Nonparametric pharmacokinetic analysis of plasma CBDCA values suggested that half-life (T1/2), area-under-the-curve and total body clearance (CLb) were not dose dependent. Volume of distribution (VDss) significantly increased with dose only between 100 and 150 mg/M2, not between 150 and 300 mg/M2. Dose-independent serum CBDCA pharmacokinetic disposition indicates that detailed investigation of tissue CBDCA distribution would be warranted and may identify novel dosing strategies that could improve the therapeutic index of CBDCA by minimizing toxicity. Clinical and laboratory findings included weight loss, hematochezia, hyperglobulinemia, hypoalbuminemia, anemia, and neutrophilic leukocytosis. Evidence of response to treatment with diminazene aceturate and ketoconazole included improvement in the abnormal clinical, hematologic, and biochemical findings, decreased serum globulin concentration and antibody titer to Leishmania donovani, and absence of organisms in examined tissues. Several foci of endemic leishmaniasis have been reported in the United States. Because of its zoonotic potential and the lack of approved treatments for dogs with leishmaniasis in the United States, the development of effective treatment strategies is needed. All dogs were fully weight bearing and free of lameness at the time of follow‐up. Radiographs were assessed for prosthetic component alignment, percentage of femoral canal fill, resorptive and formative bony changes, and changes at the implant bone interface. No evidence of infection, loosening, or failure of component fixation was seen. Settling of the acetabular component and subsidence of the femoral stem was seen early after surgery. Cortical atrophy was seen in all femurs and was the most significant bony remodeling change. Early results of clinical and radiographic evaluation of uncemented total hip arthroplasty in dogs have been encouraging. In 1 to 2 and 5 to 6 day old normal foals, median plasma endotoxin concentrations were 2.17 (range, 1.61-2.54; n = 6) and 2.89 (range, 2.61-3.50; n = 7) endotoxin units/mL (EU/mL), respectively. Median plasma endotoxin concentration in potentially septic foals with negative blood cultures or gram positive isolates (n = 8) was 2.73 (range, 0.59-4.04) EU/mL. In hospitalized foals with gram negative isolates (n = 6), median plasma endotoxin concentration was 78.06 (range, 0.76-2,696.41) EU/mL, but individual endotoxin values were only increased in foals that were extremely sick and died within hours of sampling. Plasma endotoxin concentrations were significantly greater in foals with sepsis scores > or = 11 compared with foals with sepsis scores < or = 10. Increased plasma endotoxin concentrations appear to predict an unfavorable outcome in septic foals, but normal endotoxin concentrations do not appear to have any predictive value. These horses were of various ages and breeds; they had clinical signs of colic for an average duration of 13.5 hours before examination. Seventeen horses had nasogastric reflux, and 15 had palpable small intestinal distention. Three horses were killed during surgery because of severe intestinal damage. Of the remaining 16 horses, 13 required intestinal resection and anastomosis. The length of incarcerated small intestine varied from 8 cm to 17.6 m. The ileum was involved in 12 cases. In one horse, the mesoduodenum was disrupted before surgery, causing intra-abdominal bleeding; incarceration of bowel was not found during surgery. The short-term (1 month) survival rate was 74% (14 of 19 cases), and the long-term survival rate was 63% (12 of 19 cases). The follow-up period was 3 months to 45 months (mean 17.2 +/- 7.2 months). Acute pancreatitis is commonly confirmed. There appears to be no sex predilection for the condition although the majority of affected dogs are middle aged and older. Lipid profiles of affected dogs reveal extreme elevations of fasting serum triglyceride (TG) concentrations ranging from 5·5 mmol/litre to over 90 mmol/litre; the hyperlipidaemia is almost exclusively attributed to chylomicron excess. To date, the reviewer's studies have not shown a correlation between TG levels and severity of signs. However, miniature schnauzers with fasting TG levels greater than 5·5 mmol/litre are considered to be at significant risk and should be further evaluated as candidates for dietary intervention.
