T2 - Plant Science Letters AB - Abstract A combination of techniques has been developed which permits purification of large amounts of DNA from plants containing high levels of tannins and brown pigments. The method uses lyophilized plant tissue, permitting storage of field collections and eliminating the need for fresh material. The tissue is disrupted by blending at 60° C. The DNA is separated by gel fractionation followed by hydroxyapatite (HAP) chromatography and the final product is concentrated by lyophilization. In this procedure all enzyme treatments and precipitation steps may be avoided.
