TOXICOLOGICAL SCIENCES, 49(1), 15–28.

By: A. Collins*, S. Sumner, S. Borghoff & M. Medinsky

author keywords: physiologically based pharmacokinetic (PBPK) model; tert-amyl methyl ether (TAME); tert-amyl alcohol (TAA)
MeSH headings : Administration, Inhalation; Animals; Glucuronates / metabolism; Liver / metabolism; Male; Models, Biological; Pentanols / toxicity; Protein Binding; Rats; Rats, Inbred F344; Statistics as Topic; Time Factors; Tosylarginine Methyl Ester / metabolism; Tosylarginine Methyl Ester / toxicity
TL;DR: A physiologically-based pharmacokinetic (PBPK) model was used to describe the disposition of TAME and its major metabolite, tert-amyl alcohol (TAA), in male Fischer-344 rats and indicated that the model incorporating nonspecific binding of TAA provided the best fit to the data.
