Barua, D., Faeder, J. R., & Haugh, J. M. (2008, January 21). Journal of Biological Chemistry, Vol. 283, pp. 7338–7345.

By: D. Barua n, J. Faeder* & J. Haugh n

MeSH headings : Animals; Humans; Kinetics; Models, Biological; Phosphatidylinositol 3-Kinases / chemistry; Phosphatidylinositol 3-Kinases / metabolism; Receptor, Platelet-Derived Growth Factor beta / chemistry; Receptor, Platelet-Derived Growth Factor beta / metabolism; Surface Plasmon Resonance / methods; src Homology Domains / physiology
topics (OpenAlex): Chronic Lymphocytic Leukemia Research; Protein Tyrosine Phosphatases; Glycosylation and Glycoproteins Research
TL;DR: To analyze the interactions between tandem SH2 domains and bisphosphorylated peptides, detailed kinetic models were formulated and applied to the phosphoinositide 3-kinase p85 regulatory subunit/platelet- derived growth factor β-receptor system and found homodimerization of full-length p85 was found to be an alternative mechanism for high avidity binding to phosphorylated platelet-derived growth factor receptors. By: D. Barua n, J. Faeder* & J. Haugh n

MeSH headings : Computer Simulation; Enzyme Activation; Intracellular Signaling Peptides and Proteins / chemistry; Intracellular Signaling Peptides and Proteins / metabolism; Kinetics; Models, Biological; Models, Chemical; Models, Molecular; Phosphorylation; Protein Conformation; Protein Structure, Tertiary; Protein Tyrosine Phosphatase, Non-Receptor Type 11; Protein Tyrosine Phosphatases / chemistry; Protein Tyrosine Phosphatases / metabolism; Protein Tyrosine Phosphatases / ultrastructure; SH2 Domain-Containing Protein Tyrosine Phosphatases; Structure-Activity Relationship
topics (OpenAlex): Protein Tyrosine Phosphatases; Galectins and Cancer Biology; Protein Kinase Regulation and GTPase Signaling
TL;DR: To the knowledge, this is the first kinetic model to fully consider the modular, multifunctional structure of a signaling protein, and the computational approach should be generally applicable to other complex intermolecular interactions. (via Semantic Scholar)
