The Hydroxyl Radical is a Critical Intermediate in the Voltammetric Detection of Hydrogen Peroxide

Journal of the American Chemical Society, 138(8), 2516–2519.

By: J. Roberts n, M. Voinov n, A. Schmidt n, T. Smirnova n & L. Sombers n

Contributors: J. Roberts n, M. Voinov n, A. Schmidt n, T. Smirnova n & L. Sombers n

MeSH headings : Electrochemical Techniques / methods; Electron Spin Resonance Spectroscopy / methods; Hydrogen Peroxide / analysis; Hydrogen Peroxide / chemistry; Hydroxyl Radical / analysis; Hydroxyl Radical / chemistry; Pyrroles / chemistry; Spin Labels
TL;DR: Voltammetry was combined with electron paramagnetic resonance spectroscopy to identify and define the role of an oxygen-centered radical liberated during the oxidation of H2O2, and it was demonstrated that the hydroxyl radical is a principal contributor to the voltammetry of H1O2 as signal is attenuated when this species is trapped. Multiple Scan Rate Voltammetry for Selective Quantification of Real-Time Enkephalin Dynamics

ANALYTICAL CHEMISTRY, 86(15), 7806–7812.

By: A. Schmidt n, L. Dunaway n, J. Roberts n, G. McCarty n & L. Sombers n

MeSH headings : Animals; Electrodes; Enkephalins / physiology; In Vitro Techniques; Male; Rats; Rats, Sprague-Dawley
TL;DR: This work has developed and characterized a novel voltammetric waveform for the selective quantification of small tyrosine-containing peptides, such as the ENKs, with rapid temporal (subsecond) and precise spatial (10s of micrometers) resolution, and provides a foundation for real-time measurements of endogenous ENK fluctuations in biological locations. Peroxygenase and Oxidase Activities of Dehaloperoxidase-Hemoglobin from Amphitrite ornata

Journal of the American Chemical Society, 136(22), 7914–7925.

By: D. Barrios n, J. D’Antonio n, N. McCombs n, J. Zhao n, S. Franzen n, A. Schmidt n, L. Sombers n, R. Ghiladi n

Contributors: D. Barrios n, . J. D'Antonio, N. McCombs n, J. Zhao n, S. Franzen n, A. Schmidt n, L. Sombers n, R. Ghiladi n

MeSH headings : Animals; Catalytic Domain; Hemoglobins / chemistry; Hemoglobins / metabolism; Indoles / chemistry; Indoles / metabolism; Mixed Function Oxygenases / metabolism; Models, Molecular; Oxidoreductases / metabolism; Oxygen / chemistry; Oxygen Consumption; Oxygen Radioisotopes; Polychaeta / enzymology; Protein Conformation
TL;DR: The marine globin dehaloperoxidase‐hemoglobin (DHP) from Amphitrite ornata was found to catalyze the H2O2‐dependent oxidation of monohaloindoles, a previously unreported class of substrate for DHP, and non‐enzymatic and anaerobic controls suggest a novel oxidase activity for D HP. Carbon Nanotube Yarn Electrodes for Enhanced Detection of Neurotransmitter Dynamics in Live Brain Tissue

ACS NANO, 7(9), 7864–7873.

By: A. Schmidt n, X. Wang n, Y. Zhu n & L. Sombers n

author keywords: FSCV; carbon fiber; microelectrode; dopamine; voltammetry
MeSH headings : Action Potentials / physiology; Animals; Brain / physiology; Conductometry / instrumentation; Equipment Design; Equipment Failure Analysis; Male; Microelectrodes; Molecular Imaging / instrumentation; Nanofibers / chemistry; Nanotubes, Carbon / chemistry; Nanotubes, Carbon / ultrastructure; Neurotransmitter Agents / metabolism; Organ Culture Techniques; Rats; Rats, Sprague-Dawley; Reproducibility of Results; Sensitivity and Specificity; Synaptic Transmission / physiology
TL;DR: The results demonstrate that the distinct structural and electronic properties of the nanotubes result in improved selectivity, sensitivity, and spatial resolution, as well as faster apparent electron transfer kinetics when compared to the conventional carbon-fiber microelectrodes typically used in vivo.
