Prediction of the Synergistic Glass Transition Temperature of Coamorphous Molecular Glasses Using Activity Coefficient Models

MOLECULAR PHARMACEUTICS, 18(9), 3439–3451.

author keywords: stable coamorphous pharmaceuticals; glass transition temperature; activity coefficient model
MeSH headings : Calorimetry, Differential Scanning; Chemistry, Pharmaceutical; Drug Compounding / methods; Drug Stability; Solubility; Transition Temperature; Vitrification
TL;DR: A robust thermodynamic approach extending the Couchman and Karasz method through the use of activity coefficient models, including the two-parameter Margules, non-random-two-liquid (NRTL), and three-suffix Redlich-Kister models are developed that successfully describe observed synergistic elevations and inflections in the Tg versus composition response of coamorphous pharmaceuticals.
