2023 journal article

G-actin diffusion is insufficient to achieve F-actin assembly in fast-treadmilling protrusions

BIOPHYSICAL JOURNAL, 122(18), 3816–3829.

By: R. Appalabhotla n, M. Butler*, J. Bear* & J. Haugh n

TL;DR: An experimental system in which cells adopt an approximately constant and symmetrical shape and exhibit fast, steady treadmilling in the thin region protruding from the cell is applied, revealing that diffusion alone cannot account for the transport of G-actin to the leading edge. 2021 journal article

Microfluidic devices fitted with "flowver" paper pumps generate steady, tunable gradients for extended observation of chemotactic cell migration

BIOMICROFLUIDICS, 15(4).

By: S. Baldwin n, S. Van Bruggen n, J. Koelbl n, R. Appalabhotla n, J. Bear* & J. Haugh n

TL;DR: This work reports on the development of a passive pumping strategy, driven by the high capillary pressure and evaporative capacity of paper, and its application to study fibroblast chemotaxis, and how the orientation of signaling through the phosphoinositide 3-kinase pathway affects the cells' sensing of and response to conducive gradients.
