Simultaneous oligonucleotide probe hybridization and immunostaining for in situ detection of Gordona species in activated sludge

FEMS Microbiology Ecology, 29(2), 129–136.

Contributors: D. Oerther*, F. De Los Reyes Iii, M. Hernandez* & L. Raskin*

TL;DR: The results suggest that the ribosomal RNA content of individual cells of Gordona in activated sludge is highly variable, and the combined use of an immunostain and an oligonucleotide probe targeting ribosomic RNA can determine the identity of single cells and provide an approximation of their activity. 1999 journal article

Interfacing phylogenetic oligonucleotide probe hybridizations with representations of microbial populations and specific growth rates in mathematical models of activated sludge processes

Water Science and Technology, 39(1), 11–20.

author keywords: activated sludge; biological nutrient removal; mechanistic mathematical models; oligonucleotide probes; ribosomal RNA
TL;DR: Correlations between concentrations of “theoretical” microbial populations predicted by mechanistic models and quantitative information obtained with oligonucleotide probe hybridizations are not yet developed, so future work should develop such correlations, while addressing the limitations of the molecular assays. A multiplex reverse transcription polymerase chain reaction method for the detection of foodborne viruses

JOURNAL OF FOOD PROTECTION, 62(10), 1210–1214.

By: S. Rosenfield n & L. Jaykus n

MeSH headings : Enterovirus / isolation & purification; Food Microbiology; Hepatovirus / isolation & purification; Humans; Norwalk virus / isolation & purification; Polymerase Chain Reaction / methods
TL;DR: With further streamlining efforts such as single tube amplification and liquid hybridization, multiplex PCR offers advantages over cell culture methodology and monoplex PCR because it allows for rapid and cost-effective detection of several human enteric viruses in a single reaction tube. Nucleic acid persistence in heat-killed Escherichia coli O157 : H7 from contaminated skim milk

JOURNAL OF FOOD PROTECTION, 62(8), 839–844.

By: J. McKillip*, L. Jaykus n & M. Drake*

MeSH headings : Animals; DNA, Bacterial / analysis; DNA, Ribosomal / analysis; Escherichia coli O157 / genetics; Food Contamination; Hot Temperature; Milk / microbiology; Polymerase Chain Reaction; RNA, Bacterial / analysis; RNA, Ribosomal, 16S / analysis; Reverse Transcriptase Polymerase Chain Reaction
TL;DR: DNA persistence was greater than that of rRNA following cell death by mild heat treatment in skim milk, and rRNA was less stable than DNA, suggesting its detection by RT-PCR may not be appropriate as an exclusive indicator of cell viability in minimally processed foods. A quantitative risk assessment for Bacillus cereus emetic disease associated with the consumption of Chinese-style rice

JOURNAL OF FOOD SAFETY, 19(3), 209–229.

By: D. McElroy n, L. Jaykus n & P. Foegeding n

TL;DR: A quantitative microbial risk assessment was undertaken to model the risk of the Bacillus cereus emetic foodborne disease syndrome associated with the consumption of contaminated Chinese-style boiled or fried rice, and revealed that therisk of illness was highly correlated with temperature abuse of cooked rice. ENVIRONMENTAL TECHNOLOGY, 20(2), 191–199.

By: M. Boufadel*, P. Reeser*, M. Suidan*, B. Wrenn*, J. Cheng n, X. Du*, T. Huang*, A. Venosa*

author keywords: bioremediation; oil; nutrient; beaches; shores
TL;DR: Beach microcosms containing heptadecane-coated sand were used to evaluate the effect of nitrate concentration on the hydrocarbon biodegradation rate and nitrogen recycling by the biomass was evidenced by the presence of microbial activity at zero influent nitrate concentrations.
