Early-life farm exposures and adult asthma and atopy in the Agricultural Lung Health Study

Journal of Allergy and Clinical Immunology, 140(1), 249–256.e14.

By: J. House*, A. Wyss*, J. Hoppin n, M. Richards*, S. Long*, D. Umbach*, P. Henneberger*, L. Beane Freeman ...

Contributors: J. House*, A. Wyss*, J. Hoppin n, M. Richards*, S. Long*, D. Umbach*, P. Henneberger*, L. Beane Freeman ...

MeSH headings : Aged; Asthma / epidemiology; Case-Control Studies; Environmental Exposure; Farms; Female; Humans; Hypersensitivity, Immediate / blood; Hypersensitivity, Immediate / epidemiology; Immunoglobulin E / blood; Iowa / epidemiology; Male; Middle Aged; North Carolina / epidemiology; Odds Ratio; Pulmonary Disease, Chronic Obstructive / epidemiology; Smoking / epidemiology
TL;DR: In a large US farming population, early‐life farm exposures, particularly maternal farming activities while pregnant, were strongly associated with reduced risk of atopy in adults, suggesting that protective associations of early‐ life farming exposures on atopy endure across the life course. Vagal innervation is required for pulmonary function phenotype in htr4 –/– mice

American Journal of Physiology - Lung Cellular and Molecular Physiology, 312(4), L520–L530.

By: J. House*, C. Nichols*, H. Li*, C. Brandenberger*, R. Virgincar*, L. DeGraff*, B. Driehuys*, D. Zeldin*, S. London*

Contributors: J. House*, C. Nichols*, H. Li*, C. Brandenberger*, R. Virgincar*, L. Degraff*, B. Driehuys*, D. Zeldin*, S. London*

MeSH headings : Animals; Genotype; In Vitro Techniques; Lung / innervation; Lung / physiology; Mice, Inbred C57BL; Muscle Contraction / physiology; Phenotype; Pulmonary Ventilation / physiology; Receptors, Serotonin, 5-HT4 / deficiency; Receptors, Serotonin, 5-HT4 / metabolism; Respiration; Respiratory Function Tests; Respiratory Hypersensitivity / physiopathology; Trachea / physiology; Vagotomy; Vagus Nerve / physiology; Vagus Nerve / surgery
TL;DR: It is reported here that the Htr4-/- phenotype in mouse is dependent on vagal innervation to the lung, and results suggest that HTR4's role in pulmonary function likely relates to neural innervation and control of breathing. (via Semantic Scholar)

Contributors: A. Henriquez*, J. House n, D. Miller*, S. Snow*, A. Fisher*, H. Ren*, M. Schladweiler*, A. Ledbetter*, F. Wright n, U. Kodavanti*

MeSH headings : Adrenal Cortex / metabolism; Adrenal Cortex / surgery; Adrenal Medulla / metabolism; Adrenal Medulla / surgery; Adrenalectomy; Animals; Corticosterone / blood; Cytokines / metabolism; Disease Models, Animal; Epinephrine / blood; Gene Expression Regulation; Inflammation Mediators / metabolism; Lung / metabolism; Lung / pathology; Lung Injury / blood; Lung Injury / chemically induced; Lung Injury / genetics; Lung Injury / prevention & control; Male; Neutrophils / metabolism; Oxidative Stress; Ozone; Pneumonia / blood; Pneumonia / chemically induced; Pneumonia / genetics; Pneumonia / pathology; Pneumonia / prevention & control; RNA, Messenger / genetics; RNA, Messenger / metabolism; Rats, Inbred WKY; Signal Transduction; Stress, Physiological / genetics; Transcription, Genetic
TL;DR: It is demonstrated that ozone‐induced lung injury and neutrophilic inflammation require the presence of circulating epinephrine and corticosterone, which transcriptionally regulates signaling mechanisms involved in this response. (via Semantic Scholar)

A pipeline for high-throughput concentration response modeling of gene expression for toxicogenomics

Frontiers in Genetics, 8(NOV).

By: J. House n, F. Grimm*, D. Jima n, Y. Zhou n, I. Rusyn* & F. Wright n

Contributors: J. House n, F. Grimm*, D. Jima n, Y. Zhou n, I. Rusyn* & F. Wright n

TL;DR: Using data from induced pluripotent stem cell-derived cardiomyocytes treated with chemicals at varying concentrations, a pipeline is made available for handling expression data generated by TempO-Seq to align reads, clean and normalize raw count data, identify differentially expressed genes, and calculate transcriptomic concentration–response points of departure.
