Oxygen Activation at the Active Site of a Fungal Lytic Polysaccharide Monooxygenase

ANGEWANDTE CHEMIE-INTERNATIONAL EDITION, 56(3), 767–770.

author keywords: copper; oxidoreductases; oxygen activation; polysaccharide monooxygenases; protein structures
MeSH headings : Catalytic Domain; Mixed Function Oxygenases / chemistry; Mixed Function Oxygenases / metabolism; Models, Molecular; Neurospora crassa / enzymology; Oxygen / chemistry; Oxygen / metabolism; Polysaccharides / chemistry; Polysaccharides / metabolism
TL;DR: High-resolution X-ray crystal structures of an enzyme from Neurospora crassa in the resting state and of a copper(II) dioxo intermediate complex formed in the absence of substrate are determined, providing a new structural description of oxygen activation by substrate free lytic polysaccharide monooxygenases. Ibuprofen loaded PLA nanofibrous scaffolds increase proliferation of human skin cells in vitro and promote healing of full thickness incision wounds in vivo

JOURNAL OF BIOMEDICAL MATERIALS RESEARCH PART B-APPLIED BIOMATERIALS, 105(2), 327–339.

By: M. Mohiti-Asli n, S. Saha n, S. Murphy*, H. Gracz n, B. Pourdeyhimi n, A. Atala*, E. Loboa n

author keywords: controlled release; drug delivery/release; inflammation; PLLA; wound healing
MeSH headings : Animals; Dermis / injuries; Dermis / metabolism; Dermis / pathology; Drug Carriers / chemistry; Drug Carriers / pharmacokinetics; Drug Carriers / pharmacology; Fibroblasts / metabolism; Fibroblasts / pathology; Humans; Ibuprofen / chemistry; Ibuprofen / pharmacokinetics; Ibuprofen / pharmacology; Keratinocytes / metabolism; Keratinocytes / pathology; Mice, Nude; Nanofibers / chemistry; Nanofibers / therapeutic use; Polyesters / chemistry; Polyesters / pharmacokinetics; Polyesters / pharmacology; Wound Healing / drug effects
TL;DR: It is found that degradable anti-inflammatory scaffolds containing 20 wt % ibuprofen promote human skin cell viability and proliferation in vitro, reduce wound contraction in vivo, and when seeded with skin cells, also enhance new blood vessel formation. (via Semantic Scholar)

Angiopellosis as an Alternative Mechanism of Cell Extravasation

STEM CELLS, 35(1), 170–180.

By: T. Allen n, D. Gracieux n, M. Talib n, D. Tokarz n, M. Hensley n, J. Cores n, A. Vandergriff n, J. Tang n ...

author keywords: Extravasation; Angiopellosis; Transmigration; Stem cell infusion; Diapedesis
MeSH headings : Animals; Blood Vessels / physiology; CD11 Antigens / metabolism; Cell Aggregation; Cell Membrane / metabolism; Cell Shape; Dogs; Extravasation of Diagnostic and Therapeutic Materials / pathology; Female; Humans; Injections; Intravital Microscopy; Male; Mesenchymal Stem Cells; Microspheres; Myocytes, Cardiac / cytology; Polymers / chemistry; Rats; Time Factors; Transendothelial and Transepithelial Migration; Zebrafish / metabolism
TL;DR: In injected cardiac and mesenchymal stem cells, the vascular wall undergoes an extensive remodeling to allow the cell to exit the lumen, while the injected cell remains distinctively passive in activity. (via Semantic Scholar)
